Affinity Ligands of a-Tocopherol
The purification and structural identification of proteins
and enzymes that bind tocopherol would be greatly simplified if there
existed a form of vitamin E that could be selectively and irreversibly
bound to any protein to which it could bind. To this end we have prepared
a collection of photoaffinity labels that will be tested against recombinant
a-TTP from humans and rats. The syntheses of
these labels have been designed such that one intermediate can be used
to complete the photoaffinity label or be attached to an inert matrix for
the preparation of a protein affinty-purification column.
